Chloroquine Phosphate is sold under the brand names Aralen, Chloroquine FNA, Dawaquin and Lariago among many others, available in the strength 250mg. This pharmaceutical medicine is classified as 4-aminoquinoline which works against the asexual form of malarial parasite and is available in the salts of phosphate, sulfate, and hydrochloride. It is mostly administered through oral routes in the form of tablets and is metabolized by the liver with elimination half-life 1-2 months. It is excreted mostly in urine , 50% or less as an unchanged drug and its elimination can be increased by acidification of urine. Its legal status around the globe is of ‘’ Prescription Only Medication’’. This drug is indicated for the treatment and prevention of malaria caused by certain malarial strains that are Plasmodium vivax, P. ovale, P. malariae but is not recommended in the malaria caused by Plasmodium falciparum as this strain is resistant to it. Other indications include amebiasis occurring outside the intestines, rheumatoid arthritis, and lupus erythematosus. It was also used in the treatment of COVID-19 initially but later in summer 2020 these studies were halted and is not recommended for this purpose nowadays. Common side effects are muscle problems, loss of appetite, diarrhea, and skin rash while serious side effects include vision problems, muscle damage, seizures, and anemia. FDA has not formally assigned any category for its use among pregnant women due to its safety profile hence it can not be used unless necessary. It is excreted into human breast milk but in small amounts, so decisions regarding its continuation should be made keeping mother’s and baby’s health in mind. This pharmaceutical medicine is known to have drug interactions with Ampicillin, Antacids, Cimetidine, Cyclosporine and Mefloquine. It is contraindicated in conditions like hypoglycemia, G6PD deficiency, hypomagnesemia, hypokalemia, low levels of white blood cells and alcoholism.
